Platelet retention in glass bead columns: adhesion to glass and subsequent platelet-platelet interactions.

In normal heparinized blood, the retention of platelets in glass bead columns was low in the first 1 or 2 ml, increasing to more than 80% by the 4th or 5th ml. Prior flushing of the columns with platelet-poor plasma or saline lowered retention in all 5 ml. Additional studies were carried out with a two-stage procedure in which a sample of blood (A) was pumped through a column, immediately flush...

Platelet retention in glass bead columns: further evidence for the importance of ADP.

Plasma of normal heparinized blood contained 0.284 pM ± SD 0.097 (ADP + ATP) with an ATP:ADP ratio of 2.5:1. Plasma from thrombocytopenic blood contained only 0.106 pM ± 0.073 (ADP + ATP). Blood with normal platelet retention released 0.234 pM ± 0.187 (ADP + ATP) during passage through a glass bead column, with an ATP:ADP ratio of 1.6:1. Significantly less was released in blood with low retenti...

Relationship of platelet retention in glass bead columns to the rate of aggregation with adenosine diphosphate and thrombin.

The initial rate of platelet aggregation induced by adenosine diphosphate and thrombin correlated with the number of platelets retained when samples of citrated blood from the same normal donors were passed through glass bead columns. The results support the view that the glass bead column method of measuring platelet ;adhesiveness' is influenced by platelet aggregation.

Comparison of platelet glass bead retention techniques in patients with clinical bleeding disorders.

Platelet glass bead retention by the Salzman and infusion pump techniques were compared in 36 control individuals, 10 patients with von Willebrand's syndrome and four patients with thrombocytopathies. No significant differences between the results of the two assays were observed. Eleven percent or less of the control individuals had diminished platelet glass bead retention. The mean percentage ...

Conversion from Platelet-Rich Plasma Platelet Production to Buffy Coat Platelet Component Production: Benefits and Limitations

Blood transfusion centers are under considerable pressure to produce platelet concentrates with high quality and safety due to the short shelf life of 3-5 days as well as possible bacterial and viral contaminations. For a variety of reasons, many Europeans have changed their methods of component production from whole blood using the North American standard platelet-rich plasma method to the buf...
